Shaping nanoparticle diffusion through biological barriers to drug delivery

چکیده

Nanoparticle drug delivery systems encounter many biological barriers, such as the extracellular matrix and mucus gels, that they must bypass to gain access target cells. A design parameter has recently gained attention is nanoparticle shape, it been shown elongated rod–shaped nanoparticles achieve higher diffusion rates through gels. However, optimal dimensions of rod-shaped enhance this effect yet be established. To systematically approach this, were synthesized by mechanically stretching 100 nm, 200 500 nm spherical nanoparticles. Transmission electron microscopy confirmed procedure yields a significant fraction rods remaining spheres could removed centrifugation. Fluorescent multiple particle tracking analysis was then used characterize in MaxGel®, model hydrogel. When dispersed MaxGel, we found exhibited greatest enhancement rate when their length far exceeds average hydrogel network size. These results further establish importance shape criterion improve nanoparticle-based systems.

Use of confocal microscopy for nanoparticle drug delivery through skin.

Confocal laser scanning microscopy (CLSM) is a well-used microscopic tool that provides valuable morphological and functional information within cells and tissues. The application of CLSM to skin and the topical penetration of nanoparticles (NP) will be addressed. First, we describe the advantages of confocal microscopy compared to other techniques and its use relative to skin research. Second,...

Barriers to drug delivery in solid tumors

Over the last decade, significant progress has been made in the field of drug delivery. The advent of engineered nanoparticles has allowed us to circumvent the initial limitations to drug delivery such as pharmacokinetics and solubility. However, in spite of significant advances to tumor targeting, an effective treatment strategy for malignant tumors still remains elusive. Tumors possess distin...

Trends in drug delivery through tissue barriers containing tight junctions

A limitation in the uptake of many drugs is the restricted permeation through tissue barriers. There are two general ways to cross barriers formed by cell layers: by transcytosis or by diffusion through the intercellular space. In the latter, tight junctions (TJs) play the decisive role in the regulation of the barrier permeability. Thus, transient modulation of TJs is a potent strategy to impr...
